The future of stem cells in paediatric diseases

The combination of chemotherapy and hematopoietic stem cells to restore the immune system in the treatment of various diseases in childhood

Since 2012, a study conducted at UMPMC Children’s Hospital in Pittsburgh (Pennsylvania, USA) has focused on the use of a new reduced-intensity conditioning regimen combined with umbilical cord blood in the treatment of various genetic non-cancerous disorders in children.

The group of researchers started from the well-established assumption that hematopoietic stem cells obtained from cord blood have proven to be extraordinarily effective in the treatment of many non-cancerous diseases in children, such as various blood disorders, immune system and metabolic disorders.

Traditionally, diseases such as these are treated with a massive dose of chemotherapy and drugs prior to stem cell transplantation in order to destroy as many mutated cells as possible and to prepare the body for transplanted stem cells. Unfortunately, chemotherapy can have very serious side effects, which in some cases are even fatal.

For this reason, doctors have started using reduced-intensity chemotherapy conditioning treatments, which has decreased morbidity and mortality rates, but has also increased the risk of transplant rejection and infection.

Dr. Paul Szabolcs’ team therefore focused on combining reduced-intensity chemotherapy conditioning treatment with umbilical cord blood stem cells. 44 children were treated with various disorders such as sickle-cell anaemia, thalassemia, Hunter syndrome, Krabbe disease, metachromatic leukodystrophy and many other immune deficiencies.

For more than 20 diseases, Dr. Szabolcs’ study was equally successful in transforming an umbilical cord blood transplant into a permanent transplant, with a significant improvement in the immune system condition of the treated patients.

Dr. Szabolcs and his team are enthusiastic about the results obtained and continue to work to ensure that this new method can spread and be applied on a large scale.
